01Assay development
Getting the stain right before counting anything
Immunohistochemistry and immunofluorescence assay development and optimisation on both FFPE and frozen sections: antibody selection and validation, antigen retrieval, titration, and the control design that lets a reviewer believe the signal. Multiplex panels where the biology needs more than one marker in the same section.
Tissue processing, embedding and microtomy, DAB and special stains, TUNEL for apoptosis, EdU incorporation for proliferation.
Imaging
Brightfield, fluorescence and confocal microscopy, and whole-slide scanning for anything that needs to be quantified across a section rather than sampled by eye.
02Quantification
Turning images into numbers that hold up
- Scoring strategy defined before the slides are read, not adjusted afterwards to fit.
- QuPath-based quantification and morphometry across whole slides, with classifiers trained and validated on held-out regions.
- Blinded read design so the person scoring does not know the group allocation.
- Tissue-specific biomarker localisation — not just how much signal, but which compartment and which cell type.
- Statistical treatment appropriate to nested image data, rather than treating every field as an independent observation.

Histological validation of disease progression and therapeutic response in cardiac and pulmonary tissue. Characterisation of tissue-specific biomarker expression in pulmonary hypertension models. Immunofluorescence and confocal validation of hormone-responsive 3D breast organoid cultures. Vascular remodelling assessment in pulmonary arterial disease.
